菠萝蜜果丝多糖提取工艺优化及结构、抗氧化活性分析Optimization of Extraction Process of Polysaccharides from Jackfruit Filum and Analysis of Structure and Antioxidant Activity

摘要(Abstract):
为使菠萝蜜果丝废弃物得到有效利用,获得具有良好加工性能的多糖,该研究采用碱提醇沉法,通过单因素实验与正交实验,优化菠萝蜜果丝多糖的提取工艺,以得到最佳提取条件。在此基础上探究菠萝蜜果丝多糖的体外抗氧化生物活性,以DPPH自由基、羟基自由基和ABTS自由基的清除率、IC_(50)值进行评价,并通过表征分析多糖结构。研究结果表明,最佳提取工艺条件为pH 12、提取时间2.5 h、料液比1∶6、提取温度55℃,在该条件下多糖得率为(7.70±0.10)%。傅里叶变换红外光谱显示具有多糖特征吸收峰;同步热分析测得分解温度在200~210℃之间,具有良好的热稳定性能。进一步分析菠萝蜜果丝多糖由9种单糖组成,其中主要是葡萄糖和果糖;重均分子量为12.29 kDa,多分散系数为29.62。体外抗氧化实验表明,在实验浓度范围内,菠萝蜜果丝多糖对DPPH自由基、羟基自由基、ABTS自由基具有良好的清除能力,计算得出IC_(50)值分别为0.891,1.700,0.157 mg/mL。该研究为菠萝蜜果丝废弃物的回收利用提供了一定的参考。
