柱前衍生高效液相色谱法检测酱醪中黄曲霉毒素的研究The Detection of Aflatoxin in Soy Sauce Mash by High Performance Liquid Chromatography with Pre-column Derivatization

摘要(Abstract):
目的:建立了一种基于柱前衍生的高效液相色谱-荧光检测法(HPLC-FLD)检测酱醪中黄曲霉毒素(AFT)的方法。方法:通过利用免疫亲和柱净化后,经三氟乙酸(TFA)柱前衍生,建立了HPLC-FLD检测酱醪中AFT的方法,并对10种酱醪样品中AFT的含量进行了检测。结果:该方法中AFB_1,AFB_2,AFG_1,AFG_2的定量检测限(LOQ)分别为0.32,0.35,1.10,0.35ng/mL,且这4种AFT在0.5~50ng/mL之间线性关系良好,方法稳定。不同类型样品的检测结果表明:该方法可有效克服样品中脂肪、蛋白质等杂质的干扰,同时可用于检测AFB_1,AFB_2,AFG_1,AFG_24种黄曲霉毒素。结论:该方法灵敏度高,简单快捷,适用于酱醪中AFT的检测。
