发酵法提纯草石蚕中水苏糖的工艺研究Study on Extraction and Purification of Stachyose from Stachys seiboibi by Fermentation Method

摘要(Abstract):
研究利用黑曲霉发酵强化对草石蚕的提取,运用高效液相色谱-示差折光检测法(HPLC-RID)对得到的水苏糖进行分析。采用单因素实验方法,探究了提取工艺中是否带渣发酵、蔗糖酶抑制剂、发酵培养方式等因素对水苏糖浓度的影响;确定提取水苏糖的最佳工艺条件。结果表明:接种量为提取液重量0.02%的黑曲霉,添加提取液重量0.01%的蔗糖酶抑制剂乙二胺四乙酸二钠(EDTA-2Na),采用静置-液体震荡两阶段培养方式进行带渣发酵培养,发酵温度为30.0℃,时间为24.0h,水苏糖纯度可达到80.43%。
