响应面法优化超高压提取蓝莓花色苷工艺及其活性研究Research on Optimization of Ultra-high Pressure Extraction Process of Anthocyanin from Blueberry by Response Surface Methodology and Its Activity

摘要(Abstract):
为分析萃取特性和优化工艺参数,利用超高压提取技术从蓝莓中提取花色苷。采用单因素和Box-Behnken组合试验,考察提取压力、保压时间、乙醇体积分数和料液比对蓝莓花色苷含量的影响,并分析花色苷萃取特性;通过MTT法和Annexin V-FITC/PI双标记染色法分别研究蓝莓花色苷提取物对HepG2细胞增殖和凋亡的影响。结果表明超高压提取蓝莓花色苷最优工艺参数组合为:提取压力187MPa、保压时间6 min、乙醇体积分数57%、料液比1∶29(g/mL),在此条件下花色苷含量为(5.16±0.12)mg/g。体外抗肿瘤活性表明蓝莓花色苷提取物可通过抑制HepG2细胞增殖和促进其凋亡两种途径来达到抗肿瘤效果。该研究结果可为天然活性成分的提取提供一种有效方法,同时可为食品添加剂提供重要的资源。
