毕赤酵母16B2菌株表达牛肉风味肽发酵条件的优化Optimization of fermentation conditions for beefy meaty peptide production by pichia pastoris 16B2

摘要(Abstract):
牛肉风味强化肽(BMP)最初是从牛肉的木瓜蛋白酶水解物中分离得到,其一级结构为:Lys-Gly-Asp-Glu-Glu-Ser-leu-Ala。BMP与食盐、MSG有较好的协同呈味的作用,并且具有很好的热稳定性,适合于食品工业生产中的热处理要求。因此,BMP有望代替MSG成为新一代风味强化剂。文章研究了由毕赤酵母胞外表达多拷贝牛肉风味肽的摇瓶发酵条件。确定最佳表达条件为:Yeast Nitrogen Base(YNB)的初始浓度为1%,诱导期pH为5.5,每24h添加1%的甲醇,诱导时间96h,多拷贝BMP的产量可达到约0.162g/L。
