沙棘籽粕抑菌肽的分离纯化及抑菌机制的初探Isolation and Purification of Antibacterial Peptides from Seabuckthorn Seed Meal and Preliminary Exploration of Antibacterial Mechanism

摘要(Abstract):
目的:分离纯化沙棘籽粕抑菌肽并对其抑菌机制进行初步探究。方法:以沙棘籽粕为原料,采用酶解法制备抑菌肽。以金黄色葡萄球菌为研究对象,以抑菌圈直径为评价标准,通过超滤、凝胶层析过滤(Sephadex G-25)、液相色谱与串联质谱联用(LC-MS/MS)对沙棘籽粕抑菌肽进行分离、纯化和肽段鉴定,并通过抑菌肽数据库(APD3)筛选出潜在抑菌肽段,结合分子对接手段分析其相互作用力和作用模式。测定抑菌肽最小抑菌浓度(MIC);结合激光共聚焦扫描显微镜、扫描电镜等方法从金黄色葡萄球菌细胞膜通透性和完整性等方面对其抑菌机制进行初步探索。结果:经超滤、凝胶层析分离后,F-Ⅲ-A组分具有较强的抑菌活性,通过LC-MS/MS结合抑菌肽数据库筛选得到8条潜在抑菌活性的肽段序列,分别为AIGVDILK、ALEACVKARNEGR、ARPAADER、IGGIGTVPVGR、ILSLSPSR、VGLAETIR、VLQLETAAGAAIR、VPTVDVSVVDLTVR。分子对接结果显示8条多肽和受体结合的分子对接能量范围为-10.026 5~-7.415 6 kcal/mol,平均结合能为-8.360 8 kcal/mol。MIC为2.5 mg/mL,激光共聚焦显微镜观察PI单通道显示抑菌肽处理组较对照组红色荧光强度增强,扫描电镜显示抑菌肽处理后菌体表面毛糙不光滑,并出现不同程度的皱缩、塌陷,说明抑菌肽破坏了金黄色葡萄球菌细胞膜、细胞壁的完整性。结论:对沙棘籽粕抑菌肽的分离纯化和抑菌机制的探究促进了沙棘籽粕活性肽的深度开发,并为制备抑菌肽产品提供了理论依据。
