黑皮鸡枞菌多糖提取工艺及抗氧化活性研究Study on the Extraction Technology and Antioxidant Activity of Polysaccharides from Oudemansiella raphanipies

摘要(Abstract):
以黑皮鸡枞菌子实体为试验材料,研究复合酶法提取黑皮鸡枞菌多糖的最佳工艺条件,并测定黑皮鸡枞菌多糖的抗氧化活性。分别考察复合酶比例、液料比、复合酶添加量、酶解温度、酶解pH、酶解时间对多糖提取率的影响,根据单因素试验结果,设计响应面试验优化提取工艺。通过测定DPPH自由基清除率、羟基自由基清除率评价黑皮鸡枞菌多糖的抗氧化活性。黑皮鸡枞菌多糖提取条件确定为:以纤维素酶∶果胶酶∶木瓜蛋白酶为4∶3∶3制备复合酶,复合酶添加量3.0%,液料比47∶1 (mL/g),酶解温度54℃,酶解pH 7.5,酶解时间73 min,此时,多糖提取率达18.75%。黑皮鸡枞菌多糖浓度为4.0 mg/mL时,DPPH自由基清除率、羟基自由基清除率分别达93.44%、47.58%,表现出较强的抗氧化活性。复合酶提取黑皮鸡枞菌多糖的方法具有较高的多糖提取率及抗氧化活性。该方法可以为黑皮鸡枞菌多糖的开发和利用提供理论依据和新的思路。
