高效液相色谱法测定乳酸菌发酵液中γ-氨基丁酸的含量Determination of γ-aminobutyric Acid in Lactic Acid Bacteria Fermented Liquid by HPLC

摘要(Abstract):
建立了高效液相色谱邻苯二甲醛(OPA)柱前衍生-紫外检测法对乳酸菌发酵液中γ-氨基丁酸的含量进行了测定。乳酸菌发酵液经离心,OPA柱前衍生后,利用Luna-C18色谱柱(250×4.6mm,5μm),以15mmol/mL醋酸钠缓冲液∶甲醇为55∶45为流动相,在流速为0.8mL/min,柱温30℃,紫外检测波长334nm的条件下进行测定。结果表明:γ-氨基丁酸在50~800μg/mL的浓度范围内线性关系良好,线性方程为y=5465.9x-5411(R2=0.9996),检测限为2.6714μg/mL,平均回收率为98.31%,RSD=2.27%。该方法操作简单、快速,分离度好,精密度高,满足痕量分析要求。
