高产Monacolin K红曲菌种的筛选及发酵条件的优化Screening of Monascus with High-yield Monacolin K and Optimization of Liquid-state Fermentation Conditions

摘要(Abstract):
为了提高Monacolin K的产量,应用30keV的N+离子注入,对紫红曲霉M2进行诱变育种。诱变剂量分别为1×10~(15),5×10~(15),1×1016,5×1016,1×1017 N+/cm2,通过构巢曲霉对峙培养、洛伐他汀抗性筛选、高效液相色谱HPLC方法筛选高产Monacolin K的优秀突变菌株,并且通过液态发酵试验优化部分培养条件。结果显示:相对原菌株M2,最佳诱变菌株11-13产Monacolin K的能力是原菌株M2的2.3倍,连续传代5次,其高产遗传形状稳定。其最佳发酵培养条件为:温度30℃,接种量10%,转速200r/min,发酵时间14天。
