一株产蓝色素放线菌筛选鉴定与性质初探Screening and Identification of a Strain of Actinomycete for Producing Blue Pigment and Primary Exploration of Its Nature

摘要(Abstract):
从黑龙江省牡丹江市寒地黑土中分离富集到一株产蓝紫色素的放线菌GS-1,构建发育树确定其菌属。使用碱提取法、超声提取法和SDS提取法获得蓝紫色素并比较3种方法蓝色素得率,对蓝紫色素的性质进行初步研究。结果表明:菌株GS-1PCR扩增产物的16S rRNA序列为1589bp,确定为放线菌门链霉菌属。SDS法提取的蓝紫色素得率最高;紫外-可见吸收光谱扫描该蓝紫色素的最大吸收波长为259nm;该色素易溶于水,不溶于乙醇、丙酮和乙酸乙酯;蓝色素对大肠杆菌和白葡萄球菌的生长有抑制作用;该色素在酸性条件下呈粉红色,在碱性条件下呈蓝色;光照时间越长,色素的稳定性越差,在溶液中存在NaCl时比较稳定,当NaCl的浓度为0.4mol/L时,对蓝紫色素的增色效果比较明显,在Na~+、K~+、Ca~(2+)、Mg~(2+)存在时对蓝色素的影响较小,在Ba~(2+)存在时色素溶液变浑浊。
