细菌类胡萝卜素提取工艺的优化研究Study on optimization of nature carotenoids extraction from Rhodopseudomonas palustris

摘要(Abstract):
研究了从沼泽红假单胞菌中提取天然类胡萝卜素的方法和影响提取效果的因素,并对类胡萝卜素的提取工艺进行了优化研究。结果表明,按以下工艺流程提取类胡萝卜素的效果最好:乙醇浸泡湿菌泥0.5h;49℃下丙酮(5mL丙酮:1g菌泥)搅拌浸提4h;转移到与丙酮等体积的乙醚中,水洗;10%的KOH甲醇溶液皂化(搅拌并加抗坏血酸钠);NaCl溶液洗涤。按照此优化工艺来提取类胡萝卜素,能够有效地分离细菌叶绿素和除去类胡萝卜素酯,并且类胡萝卜素产量由优化前的1.312mg/g(以湿重计)提高到1.872mg/g。
